Multi-omics AI map exposes trametinib-sensitive vulnerabilities in MSS RAS-mutant colorectal cancer
AI integration of multi-omic data from 2,000 MSS colorectal cancers identified actionable vulnerabilities and supported trametinib-based combinations in laboratory models. The work remains preclinical and requires prospective biomarker-driven trials before clinical adoption. Evidence quality is moderate: large observational dataset with functional validation but no randomized human data.
The study combined 2,000 patient samples across European cohorts with multi-omic profiling and AI-driven network analysis rather than isolated genomic or transcriptomic views. This revealed previously undetected links between RAS-mutant signaling, metabolic reprogramming and stromal interactions that standard bulk sequencing had missed. Laboratory validation in patient-derived models demonstrated that trametinib plus selected partners reduced tumor growth and extended survival, consistent with patterns seen in melanoma and lung cancer where MEK inhibition is already approved.
RCSI-ICR team: First-in-human safety run of trametinib combination in biomarker-selected MSS RAS-mutant mCRC begins within 18 months and enrolls at least 15 patients by month 24.
