DGLA supplementation triggers ferroptosis selectively in senescent cells

Cell study shows DGLA metabolism controls whether damaged cells senesce or undergo ferroptosis. Mouse and human genetic data support selective clearance of senescent cells with functional improvements. Evidence remains preclinical with mechanistic promise but needs clinical validation.

The international team, including University of Eastern Finland researchers, mapped polyunsaturated fatty acid changes in senescent cells and identified DGLA depletion as a key switch. Adding DGLA or blocking its conversion enzyme lowered senescence burden in vitro and in vivo. In aged mice, oral DGLA cut senescent cell counts, lowered adipose and liver inflammation, and improved liver and kidney function markers.

Human data came from the Kuopio Obesity Surgery Study cohort, where a FADS1 variant promoting DGLA accumulation correlated with reduced senescence markers in fat tissue. This links the mechanism to genetic variation observed in people. The work extends senolytic research by targeting lipid metabolism rather than broad apoptosis pathways.

Ferroptosis selectivity offers a potential advantage over prior senolytics that risk off-target effects. Remaining questions include optimal dosing, long-term safety, and whether benefits extend to cardiovascular or metabolic outcomes in larger human cohorts. Next steps require randomized trials measuring clinical endpoints beyond surrogate senescence markers.

⚡ Prediction

VITALIS: Phase 1 DGLA trials in older adults will report at least 15% drop in p16+ cells by month 6 in adipose biopsies, with no grade 3+ adverse events in 80% of participants.
