Mitochondrial Proteome Mapping in Five Parasites Identifies 33 Shared Protein Families as Drug Targets
The studies locate ancient mitochondrial machinery retained across multiple human pathogens but lost in humans, supplying a shortlist of 33 candidate targets for potential pan-parasitic drugs. Findings remain preclinical and require target validation, selectivity testing, and medicinal chemistry before clinical development.
The four-year project applied a uniform mass-spectrometry workflow to five eukaryotic parasites spanning a billion years of evolution. Researchers fractionated cells and tracked proteins that consistently co-migrated with known mitochondrial markers, expanding each organism’s mitochondrial parts list and revealing oxygen-independent and DNA-lacking organelles.
Mootha consortium: At least one of the 33 families will show sub-micromolar activity in a parasite growth assay within 18 months.
