Human Tau Seeds from AD and CBD Induce Exact Structural Copies in Mouse Tau over 9-12 Months
A 2026 Nature cryo-EM study shows that AD and CBD tau seeds template identical misfolded structures in mouse tau, confirming prion-like propagation at atomic resolution. The work provides mechanistic support for structure-selective therapies but is confined to rodent models and two tauopathies. Future primate studies and conformation-specific antibody trials will test translational relevance.
The findings strengthen the rationale for conformation-specific immunotherapies now in phase 2 trials. Next required experiments include longitudinal PET imaging in primates and testing whether antibodies raised against the AD versus CBD folds selectively block their respective propagation pathways.
Nature Medicine: By month 24, at least one conformation-selective anti-tau antibody will report >30% slowing of tau PET signal in a phase 2 AD cohort.
