Nanobody cocktail neutralizes multiple Indian cobra venoms in mice, offering potential antivenom advance

Preclinical mouse data demonstrate that a nanobody cocktail neutralizes Indian cobra venoms even when dosed after bite simulation. The approach addresses limitations of equine antivenom but covers only elapid neurotoxins and requires human trials. Evidence remains at the animal-model stage with clear translational gaps.

The study tested nanobodies originally raised against African mamba and cobra toxins for cross-reactivity with Indian cobra venoms. Researchers mixed five nanobodies and administered them to mice either with venom or 20 minutes later via intravenous injection. All treated mice survived without paralysis or respiratory failure, while untreated controls died within 30 minutes. The nanobodies also showed thermal stability and no binding to human proteins.

India records roughly 58,000 snakebite deaths annually, half the global total, with most victims being rural farmers and children. Current equine antivenom targets only the big four species and frequently fails against regional venom variants, while carrying risks of anaphylaxis and batch inconsistency. Nanobodies produced in microbes could reduce cost and cold-chain dependence.

The work builds on earlier nanobody successes against African elapids but remains limited to cobra venoms; it showed no activity against krait or viper toxins. Observational data indicate that delays in reaching clinics often exceed the 20-minute window tested here.

Next steps require GMP manufacturing, pharmacokinetic scaling, and phase I safety trials in humans, which have not yet been initiated for this cocktail.
