Isovalerate restores intestinal barrier function in malnourished male mice via microbiota-dependent mechanisms

A PNAS mouse study identifies isovalerate as a microbiota metabolite whose depletion contributes to barrier failure in malnutrition. Supplementation restored function in males but effects were absent in germ-free animals. Evidence is limited to surrogates in rodents; human efficacy and safety data are absent.

The work extends prior observational links between malnutrition, dysbiosis, and invasive infection but remains at the preclinical stage. Next steps require pharmacokinetic studies of isovalerate in humans, dose-finding trials, and ultimately RCTs measuring sepsis incidence and mortality in children.
