Obesity Reduces Mammary Adipocyte 9S-HODE Secretion, Removing Ferroptosis Protection Against Breast Cancer

Lean mammary adipocytes secrete 9S-HODE that induces ferroptosis in breast cancer cells; obesity enlarges adipocytes and sharply reduces this lipid, removing a natural brake on tumor growth. The Science preclinical study using mouse models and human tissue identifies a loss-of-protection pathway rather than solely a gain-of-risk pathway. Next research must test 9S-HODE restoration in clinical settings.

Huntsman Cancer Institute investigators examined mammary adipocytes from lean and obese mouse models plus human donor tissue. They measured oxylipin profiles and applied ferroptosis inhibitors to isolate the contribution of 9S-HODE. Lean adipocytes produced substantially higher levels of the lipid, increasing cancer-cell death rates; obese adipocytes showed the opposite pattern. The study reframes obesity not only as a promoter of tumor growth through inflammation and hormones but also as the removal of an endogenous restraint. This dual mechanism helps explain why weight gain elevates risk even in the absence of other metabolic derangements and why restoring the missing lipid suppressed tumors in obese mice. Because the work remains limited to preclinical systems, translation will require pharmacokinetic studies of 9S-HODE delivery, confirmation of breast-specific versus systemic production, and prospective cohorts that track 9S-HODE levels against incidence. If these steps succeed, supplementation could become a targeted adjunct for obese patients.
