Prenatal Paracetamol Linked to 40% Smaller Ovarian Volume in 3-Month-Old Girls
Observational data from two Danish cohorts associate prenatal paracetamol with reduced ovarian reserve markers in infancy and adolescence. Effects were largest with early exposure but causation remains unproven. Larger prospective studies with objective exposure biomarkers are required before clinical recommendations shift.
Confirmation requires longitudinal follow-up of the COPANA cohort through menarche and reproductive attempts, plus RCTs of non-paracetamol analgesics in pregnancy. Current data do not justify changing first-line use for short courses but underscore the need for trimester-specific exposure records in future birth registries.
VITALIS: Within 36 months, at least one registry study will report no difference in time-to-pregnancy among women with documented first-trimester paracetamol exposure versus matched controls.
