Phase 2 EVITA Trial Finds Exploratory Survival Signal With Daily Oral Vitamin C in CCUS and MDS

The EVITA phase 2 trial provides the first randomized evidence that correcting vitamin C deficiency in CCUS/MDS alters immune signaling and yields an exploratory survival benefit. The finding supports the cancer-interception strategy but requires confirmation in a larger phase 3 trial before clinical adoption. Epigenetic restoration via TET enzymes offers a plausible mechanism that warrants targeted follow-up studies.

Investigators have already begun designing a multicenter phase 3 study that will stratify by TET2 and IDH mutation status and incorporate serial single-cell epigenetic profiling to test whether the observed survival signal is mechanistically linked to restored hydroxymethylation.

⚡ Prediction

Grønbæk: A phase 3 trial enrolling 400 patients will detect a 15 % absolute improvement in three-year overall survival by 2029.
