LPC70 from soy modulates prostaglandin signaling to blunt high-salt diet effects on blood pressure and cognition in mice

Mouse experiments demonstrate that LPC70 counters high-salt-induced hypertension and cognitive impairment by rebalancing prostaglandin signaling across brain and kidney. The work highlights a potential dietary adjunct to existing sodium-reduction strategies but remains limited to short-term rodent data. Human trials are required to establish safety, dosing, and clinical endpoints.

The team fed male mice an 8% NaCl diet for 12 weeks while adding LPC70 at 0.3% w/w. They tracked tail-cuff blood pressure weekly, ran novel-object recognition and three-chamber sociability tests, and quantified prostaglandin metabolites plus receptor mRNA in cortex, hippocampus, and kidney cortex. High salt raised systolic pressure 25 mmHg, impaired recognition memory by 35%, and cut social time 28%; LPC70 normalized pressure, restored memory scores, and preserved social interaction without altering locomotion or anxiety measures.

These results extend earlier mouse work on LPC70 by mapping differential PG-EP receptor shifts: cortical PGE2 dropped while renal PGI2 rose, suggesting coordinated vascular and neuronal protection. Public-health salt-reduction campaigns have stalled at population level; a food-derived compound that targets both cardiovascular and cognitive endpoints could complement policy if human bioavailability and dosing are confirmed. The study did not test chronic dosing beyond 12 weeks or measure long-term neurodegeneration markers.

Human translation remains the critical gap. Existing observational data link urinary sodium >3.5 g/day to 1.5-fold higher dementia risk, yet no RCT has tested LPC70 or analogous lysolecithins in hypertensive or cognitively at-risk adults. A 6-month crossover trial measuring ambulatory BP, CSF biomarkers, and cognitive batteries would supply the missing causal link.

Next steps include GMP-scale LPC70 production and pharmacokinetic studies in older adults already on salt-restricted diets to define the exposure needed for measurable PG modulation.
