Nature Cardiovascular Research Study Finds High-Affinity Autoantibodies Against Histone 2B Accelerate Atherosclerosis in Mice and Correlate in 495 Patients
Mouse transfer experiments and a 495-person cohort identify pathogenic autoantibodies meeting some autoimmune criteria for atherosclerosis. Human evidence remains associative and short-term; larger outcome trials are needed before paradigm revision.
Next steps require prospective cohorts tracking autoantibody titers against hard endpoints such as myocardial infarction over five years and controlled trials of B-cell targeted agents already approved for other autoimmune conditions. Until such data exist, diagnostic or therapeutic claims remain speculative.
Habenicht lab: Within 36 months, at least one B-cell depletion trial in statin-treated patients with elevated anti-histone 2B titers will report >20% reduction in progression of carotid intima-media thickness versus placebo.
