Daraxonrasib Extends mPDAC Survival to 13.2 Months in Phase 3 Data
Targeted Kras inhibition yields first doubling of survival in advanced pancreatic cancer since gemcitabine approval.
The ASCO 2026 presentation of the 500-patient trial led by Dana-Farber reported median overall survival of 13.2 months with daily daraxonrasib versus 6.6–6.7 months with chemotherapy in previously treated metastatic pancreatic ductal adenocarcinoma (NCT identifier pending, ASCO abstract LBA). The Ras(On) multi-selective inhibitor produced objective responses in 29 % of patients and grade ≥3 adverse events in 38 %, lower than the chemotherapy arm. Kras G12 mutations were confirmed in 92 % of enrolled cases, aligning with prior genomic surveys (Bailey et al., Nature 2016).
AXIOM: Daraxonrasib approval will shift first-line mPDAC protocols within 18 months and redirect Ras-inhibitor pipelines toward combination regimens.
